Vibration issues in a 2000 GMC Sierra 2500

I have a 2000 GMC Sierra 2500 6.0 with about 146000 miles. As I'm driving down the road it will sometimes have a vibration type noise. it feels like I'm driving on the groves or notches on the side of the road. once I let off the gas it stops. it mainly happens when its in overdrive an sometimes happens while in 3rd but not as much. I have had the tires checked and rebalanced and I have also checked all the u joints and they are fine. I cannot pinpoint where it is coming from because I cant really feel it, its more of a noise type issue. I have been told it could be the overdrive module or maybe the transmission going out. Could it be that it just needs to be put on the computer and readjusted? any help will be appreciated.
 
I'd check my transmissions, could be time to change the fluid, or they might actually have issues. Even if it's shifting fine, if there's noise or vibrations it could be a sign. Anyway, it's still better to have an experienced technician check it. Hope it gets fixed soon.
